Protein Production Facility

Introduction

The Manchester Protein Production Facility (MPPF) is a state-of-the-art facility located in a purpose-built laboratory on the ground floor of the MIB. The MPPF has been established to generate clones of selected genes and/or proteins from selected expression constructs at low cost and with high efficiency. A basic service will include purification to >80% via single-step poly (His) tag-Ni 2+ affinity chromatography, but additional polishing steps will also be available upon request. The MPPF aims to promote and facilitate high throughput cloning, protein production and purification among the academic community in and around Manchester. In addition to the equipment discussed below, the MPPF has a full range of equipment for cell growth (temperature controlled shaking incubation), cell disruption (Constant Systems Cell Breaker), high speed centrifugation, electrophoresis (DNA and protein) and PCR (Biorad C100 controller and S1000 slave unit).

Protein Expression

gThe core facility provides a comprehensive resource for high level expression and scale-up production of recombinant proteins. After an initial consultation, a small scale test expression project will be carried out to optimise conditions for maximum protein solubility, yield & integrity.

We work closely with investigators at this point to discuss an appropriate scale up strategy and arrange a delivery time schedule. Initially investigators need only to provide a small aliquot of sequence verified construct. All constructs will be independently sequence verified by the facility before commencing the project. A limited molecular biology cloning service is available under special circumstances. Depending on particular needs we are able to provide quantities ranging from biochemical analysis to structural studies.

Proteomic Service

MIB will continue to offer a Proteomic Service validating recombinant proteins. The current charging system will remain and the Proteomic  Service will be administered under the umbrella of the Protein Expression Facility.
